Lacuna tool finds hidden protein pockets for small‑molecule binding

In a preprint published on 18 August, Clayton Moor described Lacuna — an open‑source tool that searches for hidden pockets where a small molecule can enter a protein.

In its baseline mode Lacuna builds 20 nearby protein states from a single structure; on one CPU core the median time per chain is 2.6 seconds. Proteins constantly shift shape, and a static model shows only one snapshot, so pockets may open only during motion.

Lacuna creates the 20 states using normal modes, finds cavities, merges overlapping ones, and ranks regions by how often they open, their volume, and their displacement.
