♻️عوارض #کم #خوابی: افزایش خطر بروز #آلزایمر و #دمانس

#Slow #wave #sleep #disruption increases #cerebrospinal #fluid #amyloid-β levels

پژوهشگران نوروساینس دانشگاه واشنگتن 🇺🇸، دانشگاه رادبوند 🇳🇱، و دانشگاه استنفورد 🇺🇸 در پژوهشی مشترک که به تازگی منتشر شده است، دریافتند حتی 1 شب #بدخوابی در افراد #میانسال #سالم منجر به افزایش چشمگیر تولید #بتاآمیلویید در #مغز می شود. همچنین، یک هفته کامل بدخوابی منجر به افزایش معنادار تولید پروتیین #تائو در مغز افراد سالم میانسال خواهد شد.
🔬در این پژوهش 17 داوطلب سالم بدون #اختلالات #خواب و بین سنین 35 تا 65 سال به مدت 5-14 روز تحت بیخوابی در مرحله خواب #nonREM قرار گرفتند و سطح بیتاآمیلویید، پروتیین تائو، #YKL-40، و #هایپوکرتین #مایع #مغزی #نخاعی (#CSF) آنها در صبح روز بعد اندازه گیری شد.
📚نتایج حاکی از آنند که اختلال در خواب با امواج آهسته به مدت یک شب منجر به افزایش معنادار بتاآمیلویید در مایع مغزی نخاعی شد. همچنین هنگامی که یک هفته مداوم این فرآیند تکرار شد، سطح پروتیئن تائو به شکل معناداری افزایش یافت.
🔆پژوهش های پیشین به ارتباط بین کم خوابی و بیماری های #عصبی-شناختی، بویژه آلزایمر و #دمانس، در #میانسالی و #سالمندی پی برده بودند. اما این پژوهش اولین مورد گزارش بالینی دلیل عصب شناختی این ارتباط محسوب می شود.

Abstract
#Sleep #deprivation increases #amyloid-β, suggesting that #chronically disrupted sleep may promote amyloid #plaques and other downstream #Alzheimer’s disease #pathologies including #tauopathy or #inflammation. To date, studies have not examined which aspect of sleep modulates amyloid-β or other Alzheimer’s disease #biomarkers. Seventeen healthy adults (age 35–65 years) without sleep disorders underwent 5–14 days of #actigraphy, followed by slow wave activity disruption during #polysomnogram, and #cerebrospinal #fluid collection the following morning for measurement of amyloid-β, #tau, #total protein, #YKL-40, and #hypocretin. Data were compared to an identical protocol, with a sham condition during polysomnogram. Specific disruption of slow wave activity correlated with an increase in amyloid-β40 (r = 0.610, P = 0.009). This effect was specific for slow wave activity, and not for sleep duration or efficiency. This effect was also specific to amyloid-β, and not total protein, tau, YKL-40, or hypocretin. Additionally, worse home sleep quality, as measured by sleep efficiency by actigraphy in the six nights preceding lumbar punctures, was associated with higher tau (r = 0.543, P = 0.045). Slow wave activity disruption increases amyloid-β levels acutely, and poorer #sleep #quality over several days increases tau. These effects are specific to neuronally-derived proteins, which suggests they are likely driven by changes in #neuronal activity during disrupted sleep.

Abstract
Sleep, a mysterious #behavior, has recently been recognized as a crucial factor for #health and #longevity. The daily sleep/wake #cycle provides the basis of #biorhythms controlling whole-body #homeostasis and #homeodynamics; therefore, #disruption of sleep causes several #physical and #psychological disorders, including #cardiovascular disease, #obesity, #diabetes, #cancer, #anxiety, #depression, and #cognitive #dysfunction. However, the mechanism linking sleep disturbances and sleep-related disorders remains unknown. #Orexin (also known as #hypocretin) is a #neuropeptide produced in the #hypothalamus. Central levels of orexin #oscillate with the #daily #rhythm and #peak at the #awake phase. Orexin plays a major role in #stabilizing the #wakefulness state. Orexin deficiency causes sleep/wake-state instability, resulting in #narcolepsy. Hyper-activation of the orexin system also causes sleep disturbances, such as #insomnia, and hence, #suvorexant, an orexin #receptor #antagonist, has been clinically used to treat insomnia. Importantly, central actions of orexin regulate #motivated behaviors, #stress response, and #energy/glucose #metabolism by coordinating the central-#autonomic #nervous systems and #endocrine systems. These multiple actions of orexin maintain #survival. However, it remains unknown whether chronopharmacological interventions targeting the orexin system ameliorate sleep-related disorders as well as sleep in humans. To understand the significance of adequate orexin action for #prevention of these disorders, this review summarizes the #physiological functions of daily orexin action and #pathological implications of its mistimed or reduced action in sleep disturbances and sleep-related disorders (#lifestyle-related physical and #neurological disorders in particular). Timed administration of drugs targeting the orexin system may prevent lifestyle-related diseases by improving the #quality of #life (#QOL) in #patients with sleep disturbances.
