هاي بروتينات كلش مهمة تفرزها الخلايا الجنينية لأنها هي المسؤولة عن تحديد الشكل العام للجسم (مثل أنّ عندك عيون 2 وإيدين 2 و midline structures وغيرها). سمّوها بإسم القنفذ hedgehog لأن شافوا أنّ لما يتعطل هذا الجين ببيوض الذباب رح تتحول إلى شكل يشبه القنفذ.

من تصير بيهن مشكلة لأي سبب كان رح يسببن تشوهات خَلقية تتراوح بين البسيطة نسبيًا مثل شفة الأرنب cleft lip & palate إلى تشوهات غريبة كلش مثل الأجنة اللي عدهم عين وحدة بس بنص وجههم cyclopia. الميكانزم اللي يشتغل بيها الـ hedgehog pathway كلش حلوة. أكو فيديو لـ Osmosis شارحيها بطريقة لطيفة تكدرون تشوفوه.
بالمناسبة هذا الـ pathway يستمر لحد البلوغ بشكلٍ أو بآخر ولما تصير بيه مشكلة ممكن يتسبب بأنواع مختلفة من السرطانات. لهذا بدأوا العلماء بتطوير أدوية تستهدفه. حاليًا أكو دوائين منهن مُرخَّصات من الـ FDA لعلاج نوع من سرطان الجلد BCC.

البشر عدهم 3 بروتينات بهذا الـ pathway ينتجها الـ notochord بالجنين، أسماءهن هنّ:

• Sonic hedgehog (Shh) protein
• Indian hedgehog(Ihh) protein
• Desert hedgehog (Dhh) protein
