لما تقرة عن الNAD+ وتكتشف انه يتكون من مادة اسمها nicotinamide اللي هي من عائلة الniacin (B3 vitamin complex) وبعدين تكتشف انه نقص الnicotinamide بالنظام الغذائي ممكن يسبب مرض اسمه pellagra اللي سببه الرئيسي هو نقص الNAD+

بس الpellagra ممكن يسببها نقص الTryptophan (حامض اميني) بالنظام الغذائي همين، لأن التريبتوفان يدخل بعملية تصنيع biosynthesis مال الNiacin اللي هو precursor للنيكوتينامايد...
بس بنفس الوقت زيادة الleucine (حامض اميني همين) ممكن تثبط عملية تحويل الniacin للnicotinamide وتسبب اعراض مشابهة للpellagra

بس...
اكو مرض ثاني هو Crohn's disease (التهاب بمناطق معينة من الامعاء) ممكن يمنع امتصاص الجسم للTryptophan او للNiacin وهم يسبب اعراض مشابهة للpellagra
