出于惯性每年还是会看看美本的录取情况。这几年真是,每年都是史上最惨烈。
今年美本不仅藤校卷,已经卷到我国人民看不上的富二代“水校”了:纽约大学总体录取率掉到了8%,南加大10%,波士顿大学11%。
要知道就在几年前,康奈尔的录取率也还有15%。
不是说好了学霸们都去高考卷清北嘛?
张一冰说:我们用十几年的时间,硬是把美本弄成了一个加强版的中国高考,一场比中国高考更加费时费力也更加残酷的马拉松比赛。这场比赛,比的不仅是参赛者的耐力体力,比的也是参赛者背后家庭的真金白银的投入,以及家长们的各显神通。

唉
可是真的有觉得自己很厉害啊
然而自己那么那么清高做出的选择,真的可以一直清高直到不后悔吗
就算是我,大概也有很会不平衡的时候吧
以上是因为”老板今天又来来research,说我们要做事情,让我不要想着time limit or technique limit or whatever, 重要的是keep the big question in your brain and keep working on it, 他也说他tried him best to not think about grant bc grant shouldn’t affect him to think about what he want to do. The principle should be you propose the great work and people fund you to do it not you published the random paper (I wanna use shitty but they are not actually all shitty but they are just random bc people don’t care and more importantly, the authors also don’t care what they published) and get money and use the money to publish more random paper. 我想起大概就上个月吧,我还专门跟老板说了这个事情,我说i really believe you can do great work that’s also why I joint the lab but we have to get money to keep the lab and then do what we wanna do. 我跟老板说: I hate the world as well and don’t like/respect most people (99%+) but the truth is we have to be adapted to their rules to get the resource and then do our work. 我跟老板说:I know what you thought and we can prove to them we are doing the best work but first we have to keep the lab!! 老板说他从来没这么想过,会想一想,几周过去了,看来老板已经完全觉得不用在乎世界是什么样的了(这才是纯粹的N吧,要是全世界都只有N应该也会很有意思吧),不管怎么样都会做自己的人其实也很有魅力啊。但是这种不被人认可的路(虽然这里的人其实也都是你也不认可的人罢了毕竟public are like this)真的是值得清高一辈子的吗。我能走多久,而老板又能走多久呢。“有感

既然都说到这了,顺便说说上个月和老板说起的其他内容
我信奉,世界充斥着无聊的人类,但是,有那么少数的,我有以我的标准非常尊敬的人,我用我的此生热爱着他们。这一生,其实就是你和这少数人的游戏,你可以recognize他们,他们也能非常精确的recognize到你,但再次之前,你要优秀,这里的优秀我指敏锐,自制,好奇,求知,然后去被他们认识,你们就可以得到互相的support和尊敬。
我的所谓的一些别人会认为是努力的部分,只是因为我有这种强烈的motivation,我想要进入这群我respect的人的圈子里,学术界如此,当然别的圈子亦有。
“我想要和我想去交流的人交流”,可能是这样的意思。

Protein Expression and Purification
CC1 was expressed from pVEX-CC1 (Quintyne et al., 1999) in BL21(DE3) cells and purified as described previously (de Cuevas et al., 1992) with minor modifications. Briefly, after overnight protein expression, cells were resuspended in Bugbuster reagent (Novagen, Madison, WI) with 25 U/ml benzonase DNase (Novagen), rocked at room temperature for 20 min, and spun at 16,000 × g for 20 min at 4°C. The supernatant was fractionated by 25% ammonium sulfate precipitation, the pellet was resuspended in phosphate buffer (16 mM Na2HPO4, 4 mM NaH2PO4, 2 mM dithiothreitol, 1 mM EDTA, 0.1 mM phenylmethylsulfonyl fluoride, pH 7.2), brought to 0.75 M KCl, and boiled in a water bath for 3 min. After clarification by centrifugation, the supernatant was loaded onto a MonoQ HR 5/5 anion exchange column (Amersham Biosciences, Piscataway, NJ) at 1 ml/min. The protein was eluted using a 0-1000 mM KCl gradient in 20 mM Tris-Cl, pH 7.0. The protein was concentrated using an Amicon centricon (3000 MW cut-off; Millipore, Bedford, MA). CC2 was expressed from pVEX-CC2 (Quintyne et al., 1999) in BL21(DE3)pLysS cells and purified as described above, except that a 50% ammonium sulfate precipitation was used.
