I've been in France for three months now. Their approach to this is simply fantastic.

I'd like to share a bit about it.

A very well known supermarket here decided to go cashless.

A group of 50 people got together and agreed that they would all do a big shop on the same day. Pilling their trolleys to the brim and each person getting to the checkout - the human check out, not the machines - and having everything scanned before bringing cash out.

The staff were absolutely overrun. There management in a complete tizz, everyone repeatedly, calmly and in a very organised and polite fashion acting as if they knew nothing. No edges, no rudeness, no humiliation.

The supermarket reinstated cash.
.....

Petrol stations : filling up with fuel then only having cash to pay.

......

Restaurants : groups of friends going out for dinner and nobody taking phones or credit cards, only cash ...

.....

They are doing it in every single area of France. Simply refusing to be part of it.

The reason it works is because it's organised in large groups. One person here and another there doesn't work. It needs to be really carefully constructed. 👏👏

CRISPR

The ability to precisely edit the genome of a living cell holds enormous potential to accelerate life science research, improve biotechnology & even treat human disease.

Methods for genome editing - primarily zinc finger nucleases & Transcription Activator-Like Effector (TALE) Nucleases -have existed for several years.. in 2013 these were quickly eclipsed by the efficiency, effectiveness & precision of the engineered CRISPR-Cas9 system.. first harnessed for mammalian genome editing by Feng Zhang of Broad Institute & MIT.

The CRISPR system

Like zinc fingers & TALEs, CRISPR systems are natural products. However CRISPR-Cas differs from zinc fingers & TALEs in one crucial aspect that makes it superior for genome editing applications: whereas zinc fingers & TALEs bind to DNA through a direct protein-DNA interaction, requiring the protein to be redesigned for each new target DNA site, CRISPR-Cas achieves target specificity through a small RNA that can easily be swapped for other RNAs targeting new sites.

In nature, CRISPR-Cas systems help bacteria defend against attacking viruses (known as bacteriophage or just phage). They consist of two components, the CRISPR (clustered, regularly interspaced palindromic repeats) array & Cas (CRISPR-associated proteins).

CRISPR sequences bookend short stretches of DNA that bacteria have copied from invading phages, preserving a memory of the viruses that have attacked them in the past. These sequences are then transcribed into short RNAs that guide Cas proteins to matching viral sequences. The Cas proteins destroy the matching viral DNA by cutting it.

Engineering the CRISPR toolbox

In early 2011 Feng Zhang..realised..a single RNA-guided protein could be a game changer in genome editing technology.. This system could target & activate genes in mammalian genomes.

Zhang & his team focused on harnessing CRISPR-Cas9 for use in human cells ..the most-cited CRISPR paper (Cong et al Science 2013).. essentially achieving “find & replace” functionality in the human genome.

In Sept 2015 Zhang & partners described a different system Cpf1 The Cpf1 system is simpler.. it requires only a single RNA. The Cpf1 enzyme is also smaller than the standard SpCas9 making it easier to deliver into cells & tissues.

The Cas9 has been modified so that instead of cutting the target DNA, it can turn gene expression on by recruiting transcriptional activators to its genomic location (Konermann et al Nature 2014).

At the Broad Institute the system has.. been used for genome-wide screening.. for cancer drugs & dissect immune regulatory networks (&) rapidly create mouse models of cancer that arise from multiple gene alterations (Platt et al Cell 2014)

The Zhang lab has trained thousands of researchers in the use of CRISPR-Cas9 genome editing technology.. & by sharing more than 40,000 CRISPR-Cas9 components.. In Sept 2015 the Zhang lab also began to share Cpf1 components.

I think those of you who know Anna DeBuisseret would all agree that she has devoted the past three years of her life to the people for the people, as many others have too. A veteran, senior lawyer and most of all a woman of heart, courage, morale integrity, grit and determination (just to name a few).
Anyone who has come across her in person or not, knows the time and effort she has invested from the goodness of her heart for humanity. Anna has worked tirelessly and unconditionally and has never taken anything and sadly, it has taken its toll on her.
We have recently learned that Anna has become seriously ill and is awaiting a diagnosis. Like many of us, Anna knows the power of natural healing and wants to go down this path and sadly has no funds to do that. This is where you can come and can make a difference to her and this cause.
Anna has become a member of our freedom family and we believe that family sticks together through thick and thin. She is a beautiful soul with a massive heart and has only ever wanted to protect people through her drive and knowledge, and she wants to continue protecting people. She cannot do that whilst she is seriously ill. We need her energy and her skill, and to do that, she needs to be well.
Donations will be used to fund all elements of her care and help her get well. If you cannot support her financially, maybe there is something else you can give at the level of your time or knowledge, just as she has done.
Anna, like many, finds it difficult to reach out for help, and when I discovered she was unwell, I encouraged her to allow me to do this for her. People want to give and we should never deny the gift of giving or deny ourselves the gift of receiving.
Please share this across all your communities, as together we can support her without placing responsibility on any one individual. No man or woman is an island and we all need support from time to time. Please give whatever you can in which every way you can. It will all be received gratefully. Let's get Anna back into this movement where she can make a difference along with all of us.
