Southern blot steps:
1- extraction of DNA
2- DNA cleaved into many fragments
3- Separation of fragments based on size by electrophoresis
4- DNA fragments in gel are denatured into ssDNA & transferred (blotted) to a nitrocellular membrane.
5- Gene of interest is only 1 or a few fragments.
6- Usage of a probe to identify the fragments of interest.
