ماهو هذا الفحص ولمن يستخدم

طبعآ هذا الفحص يستخدم للكشف عن
Total coliform و E.coli
الموجوده بالماء ..
طريقة عمل الفحص : يتم خلط 100 مل من الماء مع ميديا خاصة ويتم وضعها في incubator لمدة 24 ساعة
طبعا هنا بيكون الماء مع الميديا عديمة اللون الي هي colourless ولكن إذا تحول اللون بعد التحضين إلى الأصفر 🟡 فهذا معناته انه coliform bacteria موجوده.
وإذا تحول إلى اللون الأزرق الفاتح🔵 (يعطينا مثل الوميض إذا سلطنا عليه ultra violet light ) فهذا يعني أنه E.coli موجوده.

🔬Microbial Examination Report:-

1⃣day 1
🔸Wet preparation :-
We seen Pus cells and Bactria
🔸Culture on B.A , M.A and Choc.A

2⃣Day 2
🔹Examination and report Culture :-
if growth or non growth
🔻on M.A non growth
🔺on B.A >

🔺large colony
🔺regular colony
🔺same if hemolysis (beta hemolysis ) clear zone around colony
🔺Same do not hemolysis with gray - white colony
🔺on Choc.A
Appear large colony
Regular colony
White colony with out hemolysis

للتأكيد على ان التحلل يتم البحث عن في طبق B.A فقط
🔹Gram stain :-
▪️نفرض انها Gram positive cocci in groups
🔸Subculture:- on M.S.A
Or other plate
🔸do catalase :-
🔖Positive result⬆️bubbles
🔖negative result non⬆️bubbles
لنفرض هنا انه إجابي catalase
🔹do coagulase :-
Positive clot
نفرض انه سلبي coagulase -ve
3⃣day 3
🔎Examination and report subculture :- (M.S.A) or other
🔺large colony
🔺yellow colony
🔺raised colony
🔺circular colony
🔺smooth colony
🔹Do antibiotic sensitivity test:-

4⃣Day 4
🔎read result of antibiotic sensitivity test like Novobiocin
🔖sensitive
🔖resistant
نفرض انه resistant
