B. cereus 👇
B. cereus was grown on urea agar induce spore formation. The endospore stain was done using malachite green as the primary stain and safranin as counter the stain.
Malachite green was applied to the smear and set over a steaming water bath for 10 min then safranin added for several minutes .
A). Endospore’s stain blue, still in the vegetative cells which stains red (parent cell).
B). The vegetative cell, stains red.
C). Spores that have been released from the vegetative .

Streak plate isolation of E. coli on MacConkey Agar grown for 24 hrs at 37 degrees.
E. coli demonstrates strong lactose fermentation indicated by the bright pink halo, bile precipitant around the colonies, and pink colony growth.

Name: C. diptheriae Morphology: club shaped in "V" or "L"
Gram Stain: Gram-Positive
Bacilli Growth Characteristics: aerobic Gray to black colonies on blood tellurite agar.
Test Profile: catalase positive. Elkek test shows toxin production by agar gel precipitin test.
Description: aerobic gram positive rods that aggregate into "Chinese letters" on gram stain and has metachromatic staining granules that are enhanced with methylene blue. Vaccine consists of a toxin of diptheria toxin.
