But, what is a grimoire?

A grimoire is defined as a book of spells and incantations used for calling forth spirits. The word “grimoire” is related to the word “grammar,” which pertains to the rules and the relationships of language. Derived from the Middle English gramere and the Old French gramaire, the root of the word is an alteration of the Latin grammatica, which we can trace back to its source from the Greek feminine of grammatikos, meaning “of letters.” It seems appropriate that this feminine derivative from the language of one of the earliest classical pantheistic civilizations has evolved to describe what is now considered by many witches to be a sacred text devoted to rituals honoring the Mother Goddess. Most of the books that are now commonly classified as grimoires, however, were written during the Middle Ages and the Renaissance. They speak of many different traditions of ceremonial magic, both sacred and profane.

Most often, today's grimoire is handwritten by individuals for their own personal use. In a grimoire, there may be specific instructions that pertain to a particular tradition, or it may contain strictly personal records intended only for the use of the individual author. Sometimes the grimoire (or sections of it) is passed down or copied from a master book. The term grimoire is generic and may be used in place of the title of the actual book, which is often kept secret. What is popularly known as the “book of shadows” is really a sort of Wiccan recipe book that includes spells and incantations, but also much more. A book of shadows may also contain dreams, poems, invocations, revelations, inspiration, and lore.
Every grimoire is essentially a book of shadows, but not every book of shadows would conform to the definition of a grimoire. A book of shadows differs from a grimoire in that while it includes elements of the grimoire, it is not necessarily an exclusively instructive tome, but more of a personal record of a spiritual journey.
Some people argue that a book of shadows more closely resembles a diary or a journal and that highly personal musings have no place in the more didactic grimoire. Purists insist that the grimoire should be entirely instructional, full of information, annotation, and practical application. Fortunately for our purposes, there exists no officially designated criterion for creating a book of shadows, nor is there a correct or incorrect way of building, blessing, and usingh your personal grimoire.

از willow tree تا آسپرین

انسان‌ها قرن‌ها از فراورده‌های بید برای درد و تب استفاده می‌کردن. شواهد تاریخی استفاده از گیاهان حاوی سالیسیلات به دوران باستان می‌رسن. منابع تاریخی حتی به مصر باستان و پزشکان یونانی اشاره می‌کنن. اون آدم‌ها نمی‌دونستن COX چیه، prostaglandin چیه یا salicin چیه.
صرفن مشاهده کرده بودن: این گیاه برای بعضی دردها و تب‌ها مفیده.
قرن هجدهم، Edward Stone، روحانی انگلیسی، آزمایشی درباره‌ی پوست بید انجام داد. متوجه شد که پوست بید برای تب، به‌خصوص چیزی که اون زمان agues می‌گفتن بهش اثر داره.
گزارش‌ش سال 1763 منتشر شد و یکی از نقاط مهم در تاریخ علمی داروهای سالیسیلات محسوب می‌شه. پژوهشگرای معاصر این کار رو از اولین شواهد تجربی قانع‌کننده درباره‌ی اثر تب‌بر پوست بید می‌دونن.
سال 1829 شیمیدان فرانسوی Henri Leroux تونست salicin را از پوست بید جدا کنه.
بعد Raffaele Piria سال 1838 کار مهمی روی تبدیل/جداسازی و شناسایی salicylic acid انجام داد.
یه مشکلی وجود داشت ولی! Salicylic acid می‌تونه اثر ضد درد، ضد التهاب و تب‌بری داشته باشه اما مصرف‌ش با تحریک دستگاه گوارش همراه هست.
این شد که Charles Frédéric Gerhardt برای اولین بار acetylsalicylic acid را سنتز کرد، هرچند این محصول اون‌موقع شکل داروی امروزی توسعه پیدا نکرد.
سال 1897 شیمیدان آلمانی Felix Hoffmann که تو شرکت Bayer روی مشتق استیل‌شده‌ی سالیسیلیک اسید کار می‌کرد، این محصول رو بیرون داد:
Acetylsalicylic acid (ASA)
همون آ س آ خودمون‌ :)
ترکیبی که با نام Aspirin تجاری شد و Bayer سال 1899 به بازار معرفی کرد.

اینجا داستان آسپرین شروع می‌شه:
برای مدت زیادی، انسان می‌دونست آسپرین درد و التهاب و تب را کاهش می‌ده اما مکانیسم دقیقش مشخص نبود.
تا اینکه 1971، John Vane و همکارانش نشون دادن که آسپرین و داروهای مشابه، مسیر تولید prostaglandins رو مهار می‌کنن.
داره پیچیده می‌شه؟

داخل غشای سلول فسفولیپیدها اسیدهای چرب دارن. یکی‌شون اینه:
Arachidonic acid (AA)
حالا وقتی سلول تحریک می‌شه یا آسیب می‌بینه، آنزیم‌هایی مثل phospholipase A₂ می‌تونن AA رو از فسفولیپیدهای غشایی آزاد کنن. این AA وارد مسیرهای مختلف می‌شه. یکی‌ش مسیر COXه.
این‌ مسیر دوتا فعالیت آنزیمی مرتبط داره:
1) COX-1
بیشتر به عملکردهای homeostatic مربوطه.‌‌ مثلن عملکرد معده، کلیه و پلاکت‌ها

2) COX-2
اغلب تو پاسخ به التهاب و cytokines افزایش پیدا می‌کنه.‌

خلاصه... این چرخه AA رو اول به PGG₂ بعد به PGH₂ تبدیل می‌کنه که PGH₂ بعدن ماده‌ی اولیه‌ چندین prostanoid مختلف می‌شه:

PGE₂
تو درد، تب و التهاب نقش داره.

TXA₂
در پلاکت‌ها، thromboxane A₂ باعث افزایش فعال‌شدن و تجمع پلاکت‌ها می‌شه.

PGI₂ (prostacyclin)
در عروق اثرات متفاوتی داره و از جمله به گشادشدن عروق و مهار تجمع پلاکتی کمک می‌کنه.


آسپرین = acetylsalicylic acid
این مولکول می‌تونه یه گروه acetyl رو به یه اسیدآمینه‌ی مشخص در COX منتقل کنه! که یعنی چرخه مختل می‌شه.‌
تو بافت ملتهب، prostaglandinهایی مثل PGE₂ تو تنظیم حساسیت گیرنده‌های درد نقش دارن.
یعنی PGE₂ خودش لزوماً «درد» ایجاد نمی‌کنه، محیط رو طوری تغییر می‌ده که پایانه‌های عصبی به محرک‌های دردناک حساس‌تر بشن.
در تب هم PGE₂ تو تنظیم نقطه‌ی تنظیم حرارتی (set point) هیپوتالاموس نقش مهمی داره.
پس وقتی تولید prostaglandin کاهش پیدا می‌کنه هم درد و تب کاهش پیدا می‌کنن هم با یه سری عوارض رو به رو می‌شیم.
حالا چرا عموم می‌گن آسپرین خون‌رو رقیق می‌کنه؟ که اشتباهه.‌
پلاکت‌ها هسته ندارن. پس وقتی aspirin، COX-1 پلاکت رو به‌طور برگشت‌ناپذیر acetylate می‌کنه (مهار می‌کنه) پلاکت نمی‌تونه مثل یک سلول هسته‌دار آنزیم جدید بسازه. پس آسپرین اثر antiplatelet (ضد پلاکتی) هم داره. یعنی لخته خونی دیرتر ساخته می‌شه که یکی از عوارض جدی هست.
(مکانیسم اثر آسپرین با ضدپلاکت‌هایی مثل هپارین فرق داره بای د وی.)‌
