for the Robert Gs ... to answer your questions about David Sinclair’s longevity protocol and his take on NAD, a form of B3, it helps to see NAD in context, what it is doing and where in the inflammation, detoxification processes. You will see compounds like resveratrol, and compounds that effect/inhibits nNOS (nitric oxide synthase) like spermidine. Let’s just say, regarding the “Lifespan” protocol, ‘your mileage may differ’ depending on your own terrain: what are your toxin burdens, nutrient levels, variants in these enzymes (genes), cellular pH, minerals, salts/sugars. There are tests (urine, saliva) to tell you this information. You can enlarge these graphics to see NAD+ specifically, as well as NOS.

Dangers of Heavy Metals & How to Do a Heavy Metal Detox
Suffering from odd symptoms like fatigue, autoimmune issues and skin irritation without knowing the cause? You might be suffering from heavy metal poisoning. Here's a detox treatment plan that anyone can do.
UP CLOSE ... inflammation around TNFa, NFkB You got your glyphosate, mycotoxins, borrelia, citrinin, oxalates, heavy metals (and suggested natural and pharma compounds to block inflammatory response). You can’t pill-pop and supplement your way out of the inflammation; you are going to have to get out the toxins, clean up diet and environment
The citations (studies, primarily on rodents) are basis of Sinclair's lifespan protocol: autophagy, blood sugar management, inflammation reduction using compounds (resveratrol, spermidine, Metformin, NMN) acting on various enzymes (NOS, SIRT) and inflammatory NFkb. https://www.science.org/doi/10.1126/science.aan2788
Variants in NOS, SIRT, FOXO, and lipid metabolism (FADS), among other genes (enzymes) will influence the whole shebang but his protocol doesn't account for bioindividuality.

hmmm! "Resveratrol supplementation does not improve metabolic function in nonobese women with normal glucose tolerance."
There goes my already very lame defense of my red wine habit.

A QUICK NOTE ABOUT SPERMIDINE. It is tightly controlled in the body because it converts into hydrogen peroxide and acrolein which is a nasty toxin. You need SAM (a methyl donor) to make it. It is a polyamine (made up of many 'amines' which is broken down by an enzyme called DAO). It is ubiquitous in the body and necessary for growth. The body can make spermidine (from arginine); you can get enough of it in your diet and from microbiota in the gut.
Here is the rub.
The biochemistry of polyamines is coordinated, tightly choreographed.
Supplementing with spermidine when you have weaknesses (variants) in the enzymes that move this compound along in its tightly controlled chemistry (methylation, acetylation, catalase, glutathione) and disturbances in gut bacteria could lead to excess toxins like acrolein (see my posts above about acrolein from.polyglycol)
