From February 2017 - Lot-to-lot Consistency, Safety, Tolerability and Immunogenicity of an Investigational Hexavalent Vaccine (HV) in US Infants " This multicenter phase III study evaluated the immune responses of three different lots of new combination hexavalent vaccine that contained DTaP(5), injectable polio, Haemophilus influenzae type b (HiB), and hepatitis B. This particular study was conducted across 73 sites in the US from May 2011 through July 2013, involving a total of 2808 healthy infants. Side effects were followed for 15 days. There were 400 babies were given either Prevnar 13, for strep infection, or RV5, for rotavirus infections 2, 4 and 6 months of age, as well as HepB at 2 and 6 months of age as the “control” – the placebo – group.
 

COMMENT: The use of combination vaccines began in response to parental concern that too many vaccines were being administered in one visit. Creating a slight of hand, the first combination vaccine, Pediarix, was FDA approved in 2002.  Pediatricians can now push parents to get only one injection – but that single shot deceptively contains three vaccines: DTaP and hepatitis B and polio vaccines. In 2008, another combination vaccine was approved, Pentacel, which contains DTaP and HiB and polio.
 
Manufacturers and the Advisory Committee on Immunization (ACIP) are apparently on a mission to create a Super Shot, a single vaccine that combines as many shots in to one injection as possible to overcome “vaccine hesitancy,” the new buzzword applied to parents who question vaccines and have concerns that their infants are getting too many shots at one time.
 
This particular new, combination vaccine contains DTaP(5), a beefed-up version of other DTaP vaccines. Multiple versions of the DTaP vaccine available from different manufacturers. The primary difference lies in the number of pertussis antigens that are in the vaccine. The DTaP(5) has five components - pertussis toxoid, filamentous hemagglutinin, pertactin, and fimbriae types 2 and 3. This essentially reconstructs the very toxic whole cell pertussis vaccine (wDTP), using pieces and parts, rather than the cell wall of the entire pertussis bacterium.
 
Serious adverse events (SAEs) included intussusception and five deaths. Again, the deaths were negated with a strike of a pen as investigators determined that “no death was related to the study vaccine.”
